Relationships between cerebral tau and glucose metabolism in cognitively normal older adults

چکیده

Background Alzheimer’s disease (AD) is characterised by the accumulation of amyloid-β and tau in brain subsequent neurodegeneration. The relationship between glucose metabolism has been elucidated AD, however two early stages AD trajectory remains elusive. Here, we examine this using fluorodeoxyglucose (FDG) flortaucipir (FTP) positron emission tomography (PET) a cohort cognitively normal older adults. Method 72 participants (mean±SD age 78.7±6.3 years old) were recruited through Harvard Ageing Brain Study had available FDG-PET, FTP-PET, structural MRI scans. Region interest analysis was utilised to determine regional uptake each PET tracer. 40 split into high-tau low-tau groups based on highest 20 lowest FTP values from medial temporal lobe excluding hippocampus (MTL (no hippocampus)). FDG compared groups, while correlations examined. Result Glucose significantly higher group than whole (p = 0.023), frontal 0.028), 0.014), parietal 0.027), MTL 0.01), hippocampus) 0.002), anterior cingulate cortex 0.045), posterior 0.023). Significant negative found (r -0.317,p 0.046), -0.383,p 0.015), -0.316,p 0.047), -0.437, p 0.005), -0.444,p 0.004), -0.320,p 0.044), -0.361,p 0.022). negatively correlated with -0.326, 0.04) -0.337,p 0.034). Conclusion High levels associated hypometabolism adults, suggesting that may cause decreased without inducing cognitive deficits. Furthermore, highlight link neurodegeneration before onset decline. association reduced supports need for research intervention be aimed at preclinical AD.

Plasma Tau Levels in Cognitively Normal Middle-Aged and Older Adults

Using an ultra-sensitive technique, an immunomagnetic reduction assay, the plasma tau level can be measured to a limit of quantification of pg/ml. In total 126 cognitively normal middle-aged and older adults (45-95 years old) were recruited. The plasma tau levels were significantly higher in the older group (aged 65-95 years) 18.14 ± 7.33 pg/ml than those in the middle-aged group (aged 45-64 ye...

Correlation of specific amyloid-β oligomers with tau in cerebrospinal fluid from cognitively normal older adults.

IMPORTANCE To improve the ability to develop treatments that prevent incipient Alzheimer disease (AD) from progressing to overt AD, it is important to understand the molecular basis of the earliest pathophysiological abnormalities and to determine how amyloid-β (Aβ) is involved very early in its pathogenesis. OBJECTIVE To investigate 2 specific Aβ oligomers, Aβ trimers and Aβ*56, in human cer...

Cerebral Amyloid Deposition and Dual-Tasking in Cognitively Normal, Mobility Unimpaired Older Adults.

Background We examined relationships between cerebral amyloid-beta (Aβ) and cognitive-gait dual-task performance in 27 cognitively normal, mobility unimpaired elders. Methods We assessed Aβ on Pittsburgh Compound B (PiB)-PET. We measured gait speed separately and while performing working-memory, response-inhibition, motor-sequencing, and phone-dialing tasks. We compared dual-task costs on gai...

Cerebral Blood Flow and Amyloid-β Interact to Affect Memory Performance in Cognitively Normal Older Adults

Cerebral blood flow (CBF) alterations and amyloid-β (Aβ) accumulation have been independently linked to cognitive deficits in older adults at risk for dementia. Less is known about how CBF and Aβ may interact to affect cognition in cognitively normal older adults. Therefore, we examined potential statistical interactions between CBF and Aβ status in regions typically affected in Alzheimer's dis...

Exercise and Alzheimer's disease biomarkers in cognitively normal older adults.

OBJECTIVE In addition to the increasingly recognized role of physical exercise in maintaining cognition, exercise may influence Alzheimer's disease (AD) pathology, as transgenic mouse studies show lowered levels of AD pathology in exercise groups. The objective of this study was to elucidate the association between exercise and AD pathology in humans using Pittsburgh compound-B (PIB), amyloid-b...
